    Detecting Sentiments from Movie Reviews by Integrating Reviewers Own Prejudice

    Presently, sentiment analysis algorithms are widely used to extract positive or negative feedback scores of various objects on the basis of the text/reviews. But, an individual may have a certain degree of biasness towards a certain product/company and hence may not objectively review the object. We try to combat this biasness problem by incorporating the positive and negative bias component in the existing sentiment score of the object. This paper proposes several algorithms for a new system of implementing individual bias in the corpus of data i.e. movie reviews in this case. Each review comment has an unadjusted sentiment score associated with it. This unadjusted score is refined to give an adjusted score using the positive and negative bias score. The bias score is calculated using certain parameters, the weightage of which has been determined by conducting a survey. We lay emphasis on the degree of biasness an individual has towards or against the review parameters for the movie reviews corpus namely actor, director and genre. We equip the system with the capability to handle various scenarios like positive inclination of the user, negative inclination of the user, presence of both positive and negative inclination of the user and neutral attitude of the user by implementing the formulae we developed

    Point-of-Care Devices for Viral Detection: COVID-19 Pandemic and Beyond

    The pandemic of COVID-19 and its widespread transmission have made us realize the importance of early, quick diagnostic tests for facilitating effective cure and management. The primary obstacles encountered were accurately distinguishing COVID-19 from other illnesses including the flu, common cold, etc. While the polymerase chain reaction technique is a robust technique for the determination of SARS-CoV-2 in patients of COVID-19, there arises a high demand for affordable, quick, user-friendly, and precise point-of-care (POC) diagnostic in therapeutic settings. The necessity for available tests with rapid outcomes spurred the advancement of POC tests that are characterized by speed, automation, and high precision and accuracy. Paper-based POC devices have gained increasing interest in recent years because of rapid, low-cost detection without requiring external instruments. At present, microfluidic paper-based analysis devices have garnered public attention and accelerated the development of such POCT for efficient multistep assays. In the current review, our focus will be on the fabrication of detection modules for SARS-CoV-2. Here, we have included a discussion on various strategies for the detection of viral moieties. The compilation of these strategies would offer comprehensive insight into the detection of the causative agent preparedness for future pandemics. We also provide a descriptive outline for paper-based diagnostic platforms, involving the determination mechanisms, as well as a commercial kit for COVID-19 as well as their outlook

    MMP9 mediates acute hyperglycemia-induced human cardiac stem cell death by upregulating apoptosis and pyroptosis in vitro

    Providing a conducive microenvironment is critical to increase survival of transplanted stem cells in regenerative therapy. Hyperglycemia promotes stem cell death impairing cardiac regeneration in the diabetic heart. Understanding the molecular mechanisms of high glucose-induced stem cell death is important for improving cardiac regeneration in diabetic patients. Matrix metalloproteinase-9 (MMP9), a collagenase, is upregulated in the diabetic heart, and ablation of MMP9 decreases infarct size in the non-diabetic myocardial infarction heart. In the present study, we aim to investigate whether MMP9 is a mediator of hyperglycemia-induced cell death in human cardiac stem cells (hCSCs) in vitro. We created MMP9−/− hCSCs to test the hypothesis that MMP9 mediates hyperglycemia-induced oxidative stress and cell death via apoptosis and pyroptosis in hCSCs, which is attenuated by the lack of MMP9. We found that hyperglycemia induced oxidative stress and increased cell death by promoting pyroptosis and apoptosis in hCSCs, which was prevented in MMP9−/− hCSCs. These findings revealed a novel intracellular role of MMP9 in mediating stem cell death and provide a platform to assess whether MMP9 inhibition could improve hCSCs survival in stem cell therapy at least in acute hyperglycemic microenvironment

    Transgenic Expression of miR-133a in the Diabetic Akita Heart Prevents Cardiac Remodeling and Cardiomyopathy

    Advanced diabetes mellitus (DM) may have both insulin resistance and deficiency (double DM) that accelerates diabetic cardiomyopathy (DMCM), a cardiac muscle disorder. Reduced cardiac miR-133a, a cardioprotective miRNA, is associated with DMCM. However, it is unclear whether increasing miR-133a levels in the double DM heart could prevent DMCM. We hypothesized that increasing cardiac levels of miR-133a could prevent DMCM in Akita, a mouse model of double DM. To test the hypothesis, we created Akita/miR-133aTg mice, a new strain of Akita where miR-133a is overexpressed in the heart, by crossbreeding male Akita with female cardiac-specific miR-133a transgenic mice. We validated Akita/miR-133aTg mice by genotyping and phenotyping (miR-133a levels in the heart). To determine whether miR-133a overexpression could prevent cardiac remodeling and cardiomyopathy, we evaluated cardiac fibrosis, hypertrophy, and dysfunction (P-V loop) in 13–15 week male WT, Akita, Akita/miR-133aTg, and miR-133aTg mice. Our results revealed that miR-133a overexpression in the Akita heart prevents DM-induced cardiac fibrosis (reduced collagen deposition), hypertrophy (decreased beta-myosin heavy chain), and impaired contractility (downregulated calcium handling protein sarco-endoplasmic reticulum-ATPase-2a). These results demonstrate that increased levels of miR-133a in the DM heart could prevent cardiac remodeling. Our P-V loop analysis showed a trend of decreased cardiac output, stroke volume, and ± dp/dt in Akita, which were blunted in Akita/miR-133aTg heart. These findings suggest that 13–15 week Akita heart undergoes adverse remodeling toward cardiomyopathy, which is prevented by miR-133a overexpression. In addition, increased cardiac miR-133a in the Akita heart did not change blood glucose levels but decreased lipid accumulation in the heart, suggesting inhibition of metabolic remodeling in the heart. Thus, miR-133a could be a promising therapeutic candidate to prevent DMCM

    Bulk Level to Individual Particle Level Chemical Composition of Atmospheric Dust Aerosols (PM5) over a Semi-Arid Urban Zone of Western India (Rajasthan)

    Mineral dust particles in the lower atmosphere may significantly influence radiative and optical budgets, along with the net chemical balance, through their interactions with ambient chemicals. Their ability to absotb/scatter incoming radiation strongly depends on their chemical composition (i.e., distribution of major crustal elements), but as yet there is no adequate regional database with regard to this for the Indian region. To create a regional database of background mineral dust from a semi-arid zone of western India, we measured the chemical composition of ambient particles (with aerodynamic diameter <= 5 mu m; PM5), collected from seven locations of Jaipur city (in the vicinity of Thar Desert; Rajasthan) at varying altitudes, during late-winter of 2012. The chemical compositions of the sampled particles at both bulk and individual levels were measured using X-ray fluorescence (XRF) and Scanning Electron Microscope equipped with Energy Dispersive X-ray (SEM-EDX) techniques, respectively. Significant differences in chemical compositions were observed among the seven sites, yet the bulk chemical compositions of the particles were broadly consistent with those of individual particles. Average elemental ratios of Mg/Al, Si/Al, K/Al, Ca/Al, Mn/Al, Fe/Al were found to be 0.44 +/- 0.22, 1.96 +/- 0.90, 0.65 +/- 0.22, 1.52 +/- 0.40, 0.84 and 1.54 +/- 1.67, respectively. We also estimated the complex refractive index (RIs at 550 nm wavelength) for the studied sites, yielding an average n and k (the real and imaginary parts of RI, respectively) of (1.56 +/- 0.03) + (6.5 +/- 4.6) x 10(-3)i, with the aerosols collected from Kukas Hill area (27.02 degrees N, 75.85 degrees E) having the highest iron (Fe) mass fraction (similar to 43%). Non-crustal elements e.g., Cu, S, C, Ag and Pb were found only in aerosols over the main city Birla Temple (Jaipur) at ground level (26.89 degrees N, 75.81 degrees E)

    Challenges and opportunities in mixed method data collection on mental health issues of health care workers during COVID-19 pandemic in India

    Background: The present paper describes the key challenges and opportunities of mixed method telephonic data collection for mental health research using field notes and the experiences of the investigators in a multicenter study in ten sites of India. The study was conducted in public and private hospitals to understand the mental health status, social stigma and coping strategies of different healthcare personnel during the COVID-19 pandemic in India.Methods: Qualitative and quantitative interviews were conducted telephonically. The experiences of data collection were noted as a field notes/diary by the data collectors and principal investigators.Results: The interviewers reported challenges such as network issues, lack of transfer of visual cues and sensitive content of data. Although the telephonic interviews present various challenges in mixed method data collection, it can be used as an alternative to face-to-face data collection using available technology.Conclusions: It is important that the investigators are well trained keeping these challenges in mind so that their capacity is built to deal with these challenges and good quality data is obtained
